Lilly Says Oral GLP-1 Foundayo Reduced Predicted Diabetes and Cardiovascular Risk in ATTAIN-1 Analysis
Eli Lilly said post-hoc analyses of its Phase 3 ATTAIN-1 trial found that Foundayo, or orforglipron, was associated with meaningful reductions in predicted long-term risk of type 2 diabetes and cardiovascular disease in adults with obesity or overweight.
At 72 weeks, the highest 17.2 mg dose was associated with an estimated 57% reduction in predicted 10-year type 2 diabetes risk versus placebo and an 18% reduction in predicted cardiovascular disease risk. The 5.5 mg and 9 mg doses were associated with estimated diabetes-risk reductions of 45% and 50%, respectively.
The analysis used established risk models rather than measuring actual future cardiovascular events or diabetes diagnoses, so the findings reflect predicted risk rather than observed clinical outcomes.
Foundayo is a once-daily oral GLP-1 receptor agonist and is already approved in the U.S. for chronic weight management in adults with obesity or certain adults with overweight and related medical conditions. Unlike peptide-based GLP-1 treatments, it is a small-molecule oral drug that can be taken without food or water restrictions.
Lilly said the results add to evidence that weight-loss therapies may provide benefits beyond body-weight reduction by improving several cardiometabolic risk factors, including blood glucose, blood pressure and cholesterol.
